Criteria for prioritizing selection of Mexican maize landrace accessions for conservation in situ or ex situ based on phylogenetic analysis

Introduction A combination of factors including migration, climate change and the encroachment growing cities are associated with extinction species a reduction in biodiversity. In Mexico maize landraces an important source germplasm that could be invaluable for future breeding programs response changing environment. Efforts to develop conservation strategies hampered by costs housing maintaining large collections. Effective criteria therefore needed identify rare genotypes danger being lost define minimal cost-effective core Methods large-scale genotyping analysis Mexican was carried out using 14 SSR markers spread across 10 chromosomes. DNA extracted from 30 individual plants each accession 3 bulks containing 220ng were prepared. Amplification reactions individually on bulk sample SSRs detected automatic sequencer. Binary scores used produce phylogenies rareness index Ri developed collection accessions. Results total 40,140 accessions obtained different states covering 47 races analyzed. Accessions Northern Central-Southern distinguished based genotype Michoacán state found distinct all other 56 covered identified alleles included allele combinations determined. Discussion The strategy employed successfully regions offers cost effective method determine collections situ or ex . easily adapted use native crop such as common bean, chili pepper tomato among others.
